I have developed experimental and computational methodologies for structural determination of biological assemblies by single particle cryogenic electron microscopy (cryoEM) towards atomic resolution for three decades. Since 2008, I have established an infrastructure facilitating the determination of more than 30 single particle structures, from which the full atom models of the protein components can be obtained without using crystallography. These structures include membrane channels, protein complexes, chaperonins, RNA, RNA-protein complexes and viruses. In addition, I have extensive experience in cryogenic electron tomography of bacteria, eukaryotic cells infected by viruses and neurons. My scientific contributions have been recognized by the elections to the USA National Academy of Sciences and the Academia Sinica in Taiwan.
